چکیده

background: the aim of this study was to identify the trichomonas vaginalis strains/haplotypes based on identifying their probable variations in asymptomatic patients referred to tabriz health centers, northwestern iran. methods: sampling was taken from 50-suspected women to t. vaginalis in northwestern iran. the obtained samples were smeared and cultured. fifty dna samples were extracted, amplified and identified by nested polymerase chain reaction and pcr-rflp of actin gene using two endonuclease enzymes: m sei and r sai. to reconfirm, the amplicons of actin gene were directly sequenced in order to identify the strains/haplotypes. results: pcr-rflp patterns, sequencing and phylogenetic analyses revealed definitely the presence of the g (n=22; 73.4%) and e (n=8; 26.6%) strains. multiple alignments findings of genotype g showed five haplotypes and two amino acid substitutions in codons 192 and 211 although, no remarkable unique haplotype was found in genotype e. conclusion: the accurate identification of t. vaginalis strains based on discrimination of their unknown haplotypes particularly those which are impacted on protein translation should be considered in parasite status, drug resistance, mixed infection with hiv and monitoring of asymptomatic trichomoniasis in the region.

Role of pH on Adhesion of Trichomonas Vaginalis Isolated from Symptomatic & Asymptomatic Women to Vaginal Epithelial Cells in Vitro

Background: Vaginal epithelium is the most important site for initial contact of the parasite infection in humans. Objective: To investigate the effect of Lactobacillus acidophilus (LA) and pH on attachment of T. vaginalis isolated from symptomatic and asymptomatic patients to vaginal epithelial cells (VECs). Methods: Following 1-4 hrs contact of parasite with VECs, wet mounts were prepared and...

Shape variations of Trichomonas vaginalis in presence of different substrates

Shape variations of strains of Trichomonas vaginalis were demonstrated by scanning electron microscopy (SEM) following inoculation on McCoy cell monolayers and after interaction with human erythrocytes. The parasites adhered to both cells and presented amoeboid forms, with pseudopodia-like extensions. Several amoeboid organisms swam freely over the cell monolayers and produced aggregates. These...

Trichomonas vaginalis: characterization of ornithine decarboxylase.

Ornithine decarboxylase (ODC), the lead enzyme in polyamine biosynthesis, was partially purified from Trichomonas vaginalis and its kinetic properties were studied. The enzyme appears to be of special significance in this anaerobic parasite, since the arginine dihydrolase pathway generates ATP as well as putrescine from arginine. ODC from T. vaginalis had a broad substrate specificity, decarbox...

Detection of Trichomonas vaginalis in prostate tissue and serostatus in patients with asymptomatic benign prostatic hyperplasia

BACKGROUND Despite a worldwide common and progressive nature of benign prostate hyperplasia (BPH) in older men, no association has been observed between a causative pathogen and other etiology so far. METHODS In this study, we investigated a causative association of Trichomonas vaginalis, a flagellate protozoan parasite, in 171 BPH cases presenting without symptoms of prostatitis at a surgica...
